Desalting of Nucleic Acids by Discontinuous Diafiltration.
Materials
NALGENE Mini-Centrifuge Filter of appropriate MWCO (see application table).
Distilled water or appropriate buffer/diluent.
Micropipette
Microcentrifuge
Method
  1. Introduce up to 500ul of sample into the challenge reservoir of the NALGENE Mini Centrifuge Filter.
  2. Centrifuge at up to 13,000g.
  3. Remove filtrate from tube.
  4. Add diluent to the retentate, to a volume not exceeding 500ul and centrifuge again.
  5. This process may be repeated with the salt removal proportional to the dilution factor.
  6. Remove insert containing sample concentrate from the tube once satisfactory salt removal has been carried out. Invert insert and place in a new centrifuge tube.
  7. Spin for 1000g x 3 min to recover nucleic acid.
  8. Remove insert and discard. Cap tube to store nucleic acid.
